Combination of preoperative CEA and CA19-9 improves prediction outcomes in patients with resectable pancreatic adenocarcinoma: results from a large follow-up cohort

Abstract: Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies with a 5-year survival rate of <7%. Carbohydrate antigen 19-9 (CA19-9) and carcinoembryonic antigen (CEA) are often used to predict the outcome of the malignancy independently. However, the joint prognostic effect of the two tumor biomarkers has not been well determined. The study assessed the joint role of preoperative CA19-9 and CEA in the prognostic prediction of resectable PDAC in a large cohort of patients. The study enrolled 4… Show more

“…Serum expression of carcinoembryonic antigen (CEA, CD66e, CEACAM5) has been known for some time to be a prognostic biomarker in PDAC, albeit with rather poor sensitivity and specificity [ 77 ]. Boonstra et al used a single chain antibody fragment (scFv) that binds to CEA and labelled it with the near-infrared fluorescent dye 800CW [ 78 ], and evaluated it in a mouse model of colorectal cancer (Fig.…”

“… 33 Moreover, plasma levels of miR-16 and miR-196a in combination with CA 19-9 have been shown to work very efficiently for improving the prognostic prediction of early PDAC. 5 Furthermore, another group of researchers observed much higher levels of circulating miR-200a, 200b, and 210 in the plasma of PDAC patients. 34 …”

“… 3 Thus, in light of the disappointing statistics in the prognosis of PDAC, there is an urgent, unmet need for development of valid, reliable biomarkers for early detection and monitoring of PDAC. The current most widely used biomarkers for PDAC are carbohydrate antigen 19-9 (CA19-9), 4 carcinoembryonic antigen, 5 and/or genetic markers such as K-RAS and p53. 6 However, these markers are neither sensitive nor specific for screening, but are used to follow known disease if they were initially elevated, and are not recommended for screening and diagnosis of early disease.…”
