Combination of LC–MS- and GC–MS-based Metabolomics to Study the Effect of Ozonated Autohemotherapy on Human Blood

Abstract: Ozonated autohemotherapy (O3-AHT) is a medical approach during which blood obtained from the patient is ozonated and injected back into the body. Despite an increasing number of evidence that O3-AHT is safe, this type of therapy remains controversial. To extend knowledge about the changes in blood evoked by O3-AHT, LC-MS- and GC-MS-based metabolic fingerprinting was used to compare plasma samples obtained from blood before and after the treatment with potentially therapeutic concentrations of ozone. The proced… Show more

“…Samples were treated and analyzed in separate randomized run by a HPLC system (1200 series, Agilent Technologies, Santa Clara, CA, USA) coupled to a QTOF (6520 Agilent Technologies) mass analyzer through an ESI source, in positive and negative ion modes, as described previously . Equipment performance and reproducibility was determined using quality control (QC) samples prepared and analyzed as previously .…”

“…QA? was Information from QC filtering is also commonly lost in the case where features are only detected in one group and are completely absent in the other and are therefore often absent from QC samples (Ciborowski et al 2012a). To reflect this situation, additional in silico features were altered in the data through deleting values in one group while maintaining the original values for the other group in a random selection of eight features.…”

“…Metabolomics reflects the function of organisms from terminal symptoms of metabolic networks and demonstrates metabolite changes of a complete system caused by interventions in a holistic context, which perfectly coincides with the holistic thinking of TCM [23], [24]. High resolution 1 H nuclear magnetic resonance (NMR) spectrometry and gas chromatography–mass spectrometry (GC-MS)-based metabolome analysis study are well-established techniques for detection of the endogenous metabolic changes in biofluids such as serum and urine [25], [26]. Because of the fundamental differences in the nature of NMR and GC-MS measurements, the results obtained using each platform contribute supplementary understanding of the complex metabolic response of rats treated with CR [27].…”
