Combination chemotherapy with adriamycin (NSC‐123127) and dimethyl triazeno imidazole carboxamide (DTIC) (NSC‐45388) in children with metastatic solid tumors

Abstract: Combination chemotherapy with adriamycin and DTIC was used in 102 evaluable patients under 15 years of age who had previously treated metastatic solid tumors. Responses, defined as 50% or more reduction in all tumor masses, occurred in 10 out of 27 patients with neuroblastoma, 3 out of 8 patients with Wilms tumor, 7 out 15 patients with Ewing sarcoma, 2 out of 6 patients with osteosarcoma, 5 out of 13 patients with rhabdomyosarcoma, and 15 out of 33 patients with miscellaneous tumors which included a patient w… Show more

“…Only since the introduction of multiagent cytostatic drug treatment have long-term cures been reported. In 1975 and 1976, reports by Pomeroy and Johnson, [50] and Cangir and coworkers [51] indicated improved outcome with schemes including doxorubicin. [15,16,49] In 1976 Jaffe and coworkers [12] reported on superior survival with a combination chemotherapy called 'VAC' [vincristine, actinomycin D (dactinomycin) and cyclophosphamide] compared with single agent therapy.…”

“…Initial investigations with single agents demonstrated an excellent correlation between the murine and the human disease (3). In addition, combinations of adriamycin/ imidazole carboxamide and cyclophosphamide/imidazole carboxamide have some antitumor effect in children with widespread neuroblastoma (14,15). However, their efficacy in the model is less than that documented for BCNU and cyclophosphamide, and thus, the latter combination should be considered for human trial.…”
