Combination chemotherapy with adriamycin andcis-diamminedichloroplatinum in patients with neoplastic diseases

Abstract: Forty-eight patients with a variety of advanced solid tumors were treated with a combination of adriamycin 50 mg/m2, and cis-diamminedichloroplatinum 50 mg/m2, every 2 to 4 weeks. Fifteen patients responded with a greater than 50% regression of measurable tumor; six with lung cancer; one, carcinoma of the breast; one, ovary; one, cervix; one, prostate; one, testis; one, maxillary sinus; and one, salivary gland, plus one patient with chemodectoma and one with adenocarcinoma of unknown primary. Responses lasted … Show more

“…Both agents alone had been effective in the treatment of this n e o p l a~m .~*~~ Based on initial data available at that time, it was demonstrated that the combination of these two agents in the treatment of various far advanced malignancies resulted in a significant number of responses with tolerable t o~i c i t y .~~~~~ Subsequent reports confirmed the above finding of efficacy with this combination in patients with advanced malignancies, without major potentiation of toxicity. 1421* 31 At the present time, we have entered 12 patients onto this pilot study; the purpose is to determine the toxicity of this chemotherapeutic regimen and its apparent efficacy in the treatment of newly diagnosed nonmetastatic osteosarcoma. No attempt is being made to compare this data statistically with results obtained from other treatment regimens; thus, we have included two patients with nonextremity primary lesions in this series.…”

“…We introduced a combination of these 2 agents, termed A & P (adriamycin and platinum) (Vogl et al, 1976), which has become the basis, by adding more drugs, for the treatment of patients with ovarian cancer, small cell lung cancer and other neoplasms. Recently, we have demonstrated that DXR has a positive inoculum effect in vitro and loses efficacy at high cell densities, whereas DDP does not (Ohnuma et al, 1986).…”

“…Adriamycin was added because of its established effectiveness in Wilms' tumor [9] and most other childhood malignancies [ 10,111 . Additionally these two drugs have been shown to be synergistic in animal tumor models [12] and have demonstrated antitumor activity in several human tumors without excessive toxicity [13,14] . The use of two drugs in combination, however, precludes their evaluation as single agents.…”
