Colour variation in red grapevines (Vitis vinifera L.): genomic organisation, expression of flavonoid 3'-hydroxylase, flavonoid 3',5'-hydroxylase genes and related metabolite profiling of red cyanidin-/blue delphinidin-based anthocyanins in berry skin

Abstract: Background: Structural genes of the phenyl-propanoid pathway which encode flavonoid 3'-and 3',5'-hydroxylases (F3'H and F3'5'H) have long been invoked to explain the biosynthesis of cyanidin-and delphinidin-based anthocyanin pigments in the socalled red cultivars of grapevine. The relative proportion of the two types of anthocyanins is largely under genetic control and determines the colour variation among red/purple/blue berry grape varieties and their corresponding wines.

“…1, 2). The continuous distribution of anthocyanins here in our study is in accord with reports by Lijavetzky et al (2006) and Castellarin et al (2006) who used molecular techniques to show that anthocyanin content in grape was a quantitative character controlled by polygenes.…”

“…If breeding wine grapes, a high tartaric acid parent should be chosen. With the development of biotechnology and a completed grape genome sequence, genetic analyses are progressing (Firoozabady and Olmo 1987;Ibáñez et al 2003;Castellarin et al 2006;Lijavetzky et al 2006;Jaillon et al 2007). However, these analyses are still difficult in grape due to a long growth period, large chromosome numbers, partial sterility of ovules, and low seed germination rates (He 1999).…”

“…Such changes in hydroxylation of the B-ring of flavonoids may be determined by the relative activities and substrate specificity of F 3 0 -H and F 3 0 5 0 -H, Castellarin et al 2006). F 3 0 5 0 -H belong to a large multigenic family in the grapevine genome (Jaillon et al 2007;Velasco et al 2007), each one being probably regulated in a precise time and tissue specific manner Jeong et al 2006).…”

“…However, the nonanthocyanin polyphenolic composition of the leaves of these varieties is unstudied. Since genetic differences among phenotypes may render the importance of environmental factors on phenolics production moot [25], these varieties offer an amenable model to determine the importance of genetic and environmental factors influencing grapevine phenolic composition. Nevertheless, the native varieties bring diversity; therefore, their characterization is critical.…”
