Colorectal carcinoma antigens detected by hybridoma antibodies

Abstract: Hybridoma cells which secrete colorectal carcinoma-specific antibodies have been produced and used to study the antigenic structure of these tumor cells. Nineteen antibodies have been studied in detail, and 15 of these are colorectal carcinoma specific. Only two antibodies reactive with carcinoembryonic antigen (CEA) have been discovered and five other antibodies that react with distinct epitopes on the cell surface have been defined. Several antigens with distinct molecular characteristics have been shown to … Show more

“…1 -4 Anti -CRC mAb CO -029, which binds to an unrelated CRC Ag, has been described. 28 Native and recombinant GA733 -2E proteins, the latter derived from recombinant baculovirus, were used in in vitro lymphocyte stimulation and enzyme -linked immunosorbent assay (ELISA ), respectively, and have been described. 20 Recombinant VV and adenovirus ( Ad ) expressing GA733 Ag The GA733 -2 cDNA was subcloned as an SmaI-PstI fragment into a plasmid under the control of the VV promoter pH5R.…”

“…In 1979, Koprowoski et al utilized hybridoma technology to identify carbohydrate antigen 19-9, a recognizable sialoganglioside first described in the colorectal cancer cell line SW1116, by using a monoclonal antibody called 1116-NS-19-9 [6]. CA 19-9 also referred to as sialyl Lewis-a (sLea), is expressed on the surface of cancer cells as a glycolipid and as an O-linked glycoprotein [7].…”

“…CA 19-9 also referred to as sialyl Lewis-a (sLea), is expressed on the surface of cancer cells as a glycolipid and as an O-linked glycoprotein [7]. Subsequently, CA 19-9 was also identified in the tissue and sera of patients with other gastrointestinal tumors including esophageal, gastric, biliary and pancreatic cancer [6,7]. CA 19-9 is derived from an aberrant pathway during production of its normal counterpart disialyl Lewis-a which has one extra sialic acid residue attached through a 2→6 linkage.…”

“…The following day, the cell media were removed, and each well was washed with 150 mL of PBS and 2% FBS. The wash was also removed, and each well was incubated for 1 h at room temperature with 150 mL of a 1 nM concentration of the parental intact anti-CA19-9 mAb obtained from purifying the supernatant of the 116-N-19-9 hybridoma cells known to produce the mAb [18]. After incubation, the wells were washed similarly and incubated for 1 h at room temperature with 150 mL of varying concentrations, ranging from 0.01 to 100 nM, of the anti-CA19-9 cys-diabody.…”
