Color Reaction of Anthrone with Monosaccharide Mixtures and Oligo- and Polysaccharides Containing Hexuronic Acids

Abstract: Synthetic mixtures of galacturonic acid, galactose, and rhamnose have been quantitatively resolved. Overall concentrations as low as 21 y per ml. were used. The color produced by the reaction of di-, tri-, or tetragalacturonic acid with anthrone obeys Beer's law, at least up to 200 y per 6 ml.; the quantity of color is pro-

“…Above 7O"C, however, all aged hydrocolloid-anthrone mixtures showed an increase in color density. Up to 24-48 hr of aging this was particularly outstanding with pectin and sodium alginate and is in agreement with previous findings (Helbert andBrown, 1956, 1957).…”

“…Influence of added inorganic salts and other excipients on the intensity of the color produced by carrageenan (Seakem type 5) in the anthrone reaction. (Helbert and Brown, 1961) . Some relationship was found between the heating period and aging and between the heating temperature and aging and their influence on color intensity.…”

“…Enough 5N NaOH was added to give a final concentration of 0.5N and the mixture was treated as shown in the outline. The carrageenan content of filtrate III was determined on a suitable aliquot by the anthrone-thiourea method (Graham, 1965) or by the barium chloride procedure (Hansen and Whitney, 1960 Brown, 1957Brown, , 1961. Prior to analysis of any of cust bean gum (neutral polysaccharide) ; any prethe fractions, it was shaken with 5 g of mercuric cipitate on the filter paper was returned to the chloride and filtered, and the filtrate was used for centrifuge tube.…”
