Matrisome-associated heparin binding proteins (HBPs) with roles in extracellular matrix assembly are strongly correlated to β-amyloid (Aβ) and tau pathology in Alzheimer’s disease (AD) brain and cerebrospinal fluid (CSF). However, it remains challenging to detect these proteins in plasma using standard mass spectrometry (MS)-based proteomic approaches. Here we utilized heparin affinity chromatography for the capture and enrichment of HBPs in plasma from healthy control and individuals with AD. This method was highly reproducible and effectively enriched well-known HBPs like APOE and thrombin, while also efficiently depleting high-abundance proteins such as albumin. To increase the depth of our analysis of the heparin-enriched plasma proteome and compare differences in disease we applied off-line fractionation and tandem mass tag mass spectrometry (TMT-MS) to compare the proteomic profiles between AD and control individuals across two datasets (n= 121 total samples). This led to the identification of 2865 proteins, spanning 10 orders of magnitude in protein abundance within the plasma. Notably, HBPs were some of the most increased proteins in AD plasma compared to controls. This included members of the matrisome-associated module in brain, SMOC1, SMOC2, SPON1, MDK, OLFML3, FRZB, GPNMB and the ɛ4 isoform of APOE. Heparin-enriched plasma proteins also exhibited strong correlations to conventional AD biomarkers including CSF Aβ, total tau (tTau), and phosphorylated tau (pTau) as well as plasma pTau supporting their role as potential surrogate markers of underlying brain pathology. Utilizing a consensus AD brain protein co-expression network, we assessed relationship between the plasma and brain proteomes and observed that specific plasma proteins exhibited consistent direction of change in both brain and plasma, whereas others displayed divergent changes, further highlighting the complex interplay between the two compartments. In summary, these findings provide support for the integration of a heparin enrichment method with MS-based proteomics for identifying a wide spectrum of plasma biomarkers that mirror pathological changes in the AD brain.

BackgroundLiver cancer (LC) is well known for its prevalence as well as its poor prognosis. The aberrant expression of lysyl oxidase (LOX) family is associated with liver cancer, but their function and prognostic value in LC remain largely unclear. This study aimed to explore the function and prognostic value of LOX family in LC through bioinformatics analysis and meta-analysis.ResultsThe expression levels of all LOX family members were significantly increased in LC. Area under the receiver operating characteristic curve (AUC) of LOXL2 was 0.946 with positive predictive value (PPV) of 0.994. LOX and LOXL3 were correlated with worse prognosis. Meta-analysis also validated effect of LOX on prognosis. Nomogram of these two genes and other predictors was also plotted. There was insufficient data from original studies to conduct meta-analysis on LOXL3. The functions of LOX family members in LC were mostly involved in extracellular and functions and structures. The expressions of LOX family members strongly correlated with various immune infiltrating cells and immunomodulators in LC.ConclusionsFor LC patients, LOXL2 may be a potential diagnostic biomarker, while LOX and LOXL3 have potential prognostic and therapeutic values. Positive correlation between LOX family and infiltration of various immune cells and immunomodulators suggests the need for exploration of their roles in the tumor microenvironment and for potential immunotherapeutic to target LOX family proteins.

Background: Distant metastasis remains the main reason for treatment failure in nasopharyngeal carcinoma (NPC) after radical treatment. In this study, we aim to identify the key genes related to the metastasis of NPC via bioinformatics analysis and immunohistochemistry verification, so as to provide a potential therapeutic biomarker of metastatic NPC. Methods: One dataset (GSE149587) from the Gene Expression Omnibus (GEO) database was analysed to screen the differentially expressed genes (DEGs) between nonmetastatic and metastatic NPC plasma. Functional enrichment analysis for DEGs was performed using an online bioinformatics tool. Protein‒protein interaction (PPI) networks were constructed via the STRING database. The hub gene was verified by immunohistochemistry (IHC) assay in NPC samples. Results: A total of 65 DEGs were screened from the dataset,with 6 downregulatedand 59 upregulatedin NPC plasma samples. Enrichment analysis demonstrated that DEGs were mainly enriched in positive regulation of cytosolic calcium ion concentration, response to hypoxia, cell surface receptor signaling pathway and calcium signaling pathway. The PPI network was established, and one significant module with 9 vital genes was identified via MCODE. Among these nine hub genes, CSF1 and CD4 had the highest node degrees. IHC staining indicated that high expression of receptor of CSF-1, CSF-1R, has a positive correlation with lymph node metastasis, distant metastasis and clinical stage. Conclusion: The present study indicated that CSF-1R was correlated with metastasis in NPC patients and might be a potential therapeutic biomarker of metastatic NPC.
