The development of food-grade carriers based on EFSA and/or FDA-approved polymeric materials is an area of growing interest for the targeted delivery of bioactive compounds to the colon. Many nutraceuticals have shown promise in the local treatment of conditions that threaten quality of life, such as ulcerative colitis, Crohn’s disease, colorectal cancer, dysbiosis and other problems affecting the gut and colon. Nevertheless, their bioavailability is often limited due to poor solubility, rapid metabolism and low permeability, as well as undesirable local side effects. Encapsulation in carriers, which can protect the active ingredient from degradation and improve absorption and targeted administration in the colon, is one way to overcome these limitations. The technological characterization of these systems is important to assess their efficacy, safety and stability. In particular, morphology, size and surface properties influence their actions and interaction with the bio-phase. Meanwhile, encapsulation efficiency, profile and in vitro release kinetics are key parameters to assess the ability to reach the target site. This paper proposes a recent review of food-grade polymer-based systems for colorectal targeting of bioactive substances, focusing on their technological characterization and assessment of stability and biological activity, which are important in determining their full bench-to-bed potential.

Inkjet printing (IJP) is an additive manufacturing process that selectively deposits ink materials, layer‐by‐layer, to create three‐dimensional (3D) objects or two‐dimensional (2D) patterns with precise control over their structure and composition. This technology has emerged as an attractive and versatile approach to address the ever‐evolving demands of personalized medicine in the healthcare industry. Although originally developed for non‐healthcare applications, IJP harnesses the potential of pharma‐inks, which are meticulously formulated inks containing drugs and pharmaceutical excipients. Delving into the formulation and components of pharma‐inks, the key to precise and adaptable material deposition enabled by IJP is unraveled. The review extends its focus to substrate materials, including paper, films, foams, lenses, and 3D‐printed materials, showcasing their diverse advantages, whilst exploring a wide spectrum of therapeutic applications. Additionally, the potential benefits of hardware and software improvements, along with artificial intelligence integration, are discussed to enhance IJP's precision and efficiency. Embracing these advancements, IJP holds immense potential to reshape traditional medicine manufacturing processes, ushering in the era of medical precision. However, further exploration and optimization are needed to fully utilize IJP's healthcare capabilities. Patchouli oil has exhibited remarkable efficacy in the treatment of colitis. However, its volatility and potential irritancy are often drawbacks when extensively used in clinical applications. Oil gel is a semisolid and thermoreversible system that has received extensive interest for its solubility enhancement, inhibition of bioactive component recrystallization, and the facilitation of controlled bioactive release. Therefore, we present a strategy to develop an oil gel formulation that addresses this multifaceted problem. Notably, a patchouli oil gel formulation was designed to solidify and trap patchouli oil into a spatially stable crystal-particle structure and colonic released delivery, which has an advantage of the stable structure and viscosity. The patchouli oil gel treatment of zebrafish with colitis improved goblet cells and decreased macrophages. Additionally, patchouli oil gel showed superior advantages for restoring the tissue barrier. Furthermore, our investigative efforts unveiled patchouli oil's influence on TRP channels, providing evidence for its potential role in mechanisms of antiinflammatory action. While the journey continues, these preliminary revelations provide a robust foundation for considering the adoption of patchouli oil gel as a pragmatic intervention for managing colitis.
