Colloidal Gold Filtrates as Metal Substrates for Surface-Enhanced Infrared Absorption Spectroscopy

Abstract: A new method for obtaining surface-enhanced infrared absorption (SEIRA) spectra of antibodies and antibody/antigen complexes has been developed. Antibodies attached to colloidal gold particles and then collected by filtration onto porous polyethylene membranes show enhanced spectral bands at 1080 and 990 cm-1 regardless of the antibody specificity. Attachment of a model antigen, glucose oxidase, to its specific antibody/colloid complex prior to collection produces enhanced bands at 1540, 1395, and 1250 cm-1. S… Show more

“…Doseresponse curves were created for spiked buffer samples to assess the MAAB's overall performance and to provide a basis for comparison with other so-called rapid methods of detection. The LODs determined here for both 15-min and 1-h protocols (ϳ10 4 CFU/ml) compare favorably with those for several other antibody-based systems (LODs in the range of mid-10 3 to 10 5 CFU/ml), including piezoelectric sensors (4,33,43,45,59), fiber optic sensors (44,71,72), an interferometer (57), surface plasmon resonance-based sensors (9,56), and an electrochemical detector (22). ORIGEN, a system based on electrochemiluminescence, has proven to be the most sensitive rapid detection system, with a detection limit of 1,000 CFU/ml (70).…”

“…[24][25][26]44] Therefore, the increase in the IR absorption relative to the smooth surface is attributed to the SEIRS effect. However, as can be seen in Figure 3, the IR absorption at approximately 1234 cm À1 actually decreases with additional NP-deposition steps (although after the first layer of Au NPs, the amount of PDT increases with the number of Au NP depositions, see Scheme 1).…”

“…Normally, immunoassay requires selective quantitation of trace levels of target molecules in complex biosystem. However, former IR immunoassays were ex situ and showed poor detection limit which extremely restricted the application [72,73]. In 2012, ATR-SEIRAS technique was first introduced into immunoassay by our group [74].…”
