Colletoic Acid, a Novel 11β-Hydroxysteroid Dehydrogenase Type 1 Inhibitor from Colletotrichum gloeosporioides SANK 21404

Abstract: Colletoic acid, a novel 11b -hydroxysteroid dehydrogenase type 1 (11b -HSD1) inhibitor, was found and isolated from the cultured broth of the producing fungus Colletotrichum gloeosporioides SANK 21404. Its structure was determined to be a novel acorene-type sesquiterpene by several spectroscopic methods. The absolute structure of colletoic acid was established using a modified Mosher's method and single-crystal X-ray diffraction analysis.

“…Colletotrichum is known for its production of the anticancer drug taxol (Gangadevi and Muthumary 2008 ). The medicinally and industrially important extra cellular lipase (Balaji and Ebenezer 2008 ), an antimicrobial metabolite (colletotric acid (Zou et al 2000 )) and colletotric acid (an 11 β -hydroxysteroid dehydrogenase type 1 (11 β -HSD1) inhibitor (Aoyagi et al 2008 )) were also isolated from these fungi genera. The cosmopolitan lifestyle of this interesting fungal group is likely a consequence of the high morphology and genetic variation that happens in the species of this genus (Sutton 1992 ).…”

“…Hexadecene was isolated from an extract of Monochaetia kansensis , Dodecene was isolated from an endophytic fungi Fusarium solani associated with Taxus baccata [ 51 ]. Dodecanoic acid ethyl ketones, Phthalic acid, octyl 2-pentyl ketones might be the reason for antimicrobial activity of endophytic fungi [ 52 ]. Neodin 18, Naphthalene is used as an antimicrobial, insecticide, insect repellent, anthelminthic and vermicide [ 53 ].…”

“…This journal is © The Royal Society of Chemistry 2015 (+)-Colletoic acid is a sesquiterpenoid also from the acorane family and a potent inhibitor of the 11β-hydroxysteroid type 1 enzyme, making it a therapeutic target for metabolic syndrome. 149 This compound was generated as a single enantiomer in 2013 by a diastereoselective intramolecular 5-exo-Heck reaction. 150 At the same time, Nakada et al used a stereoselective Lewis-acid catalysed Diels-Alder reaction with isoprene to create the spirocentre 151,152 (Scheme 48).…”
