Collagenolytic Activity of Crevicular Fluid and of Adjacent Gingival Tissue

Abstract: The fluid in and gingival tissue lining periodontal pockets were collected from male patients undergoing periodontal therapy. The collagenolytic activity of the crevicular fluid, and the ability of the gingiva in culture to degrade (a) an exogenous collagen substrate, and (b) endogenous collagen newly synthesized and labeled with H3-hydroxyproline, were related to the severity of gingival inflammation. Although inflammation appeared to have only a slight effect on gingival collagenolytic activity and on the tu… Show more

“…In the present study, MMP-9 demonstrates a high gingivitis/healthy and periodontitis/healthy ratio but only the former ratio passed the prescribed criteria. Its activation by periodontopathic bacteria (93) is supported by literature that establishes elevated levels in diseased gingiva (33,94) and gingival crevicular fluid (42,95,96) where evidence of more active collagenolytic activity and inflammation is seen (97). MMP-9 was 3.2 times and 4.6 times greater in the gingivitis and chronic periodontitis groups respectively when compared to the healthy group in our study.…”

“…Perhaps the most surprising finding in the experiments summarized above was the lack of convincing association between collagenase activity and GI score in patients other than those with LJP. This is clearly at variance with the results of previous studies for reasons that are not quite clear (3,4). It is still uncertain what aspect of the disease each of these parameters measures and how they relate, if at all, to disease progression.…”

“…Evidence has been accunaulated that interstitial collagenase of human cellular origin is of primary itnportance in the initiation of destruction of gingival coUagens (4)(5)(6). Collagenase has been shown to be present in inflamed human gingiva and the activity of collagenase in gingival tissue and crevicular fluid has been shown to correlate with parameters of chronic gingival inflammation (7)(8)(9)(10)(11)(12)(13). In addition to collagenase, the presence of other neutral proteolytic enzymes in inflamed human gingiva has been demonstrated (9,14).…”
