Collagen remodelling in the guinea-pig uterine cervix at term is associated with a decrease in progesterone receptor expression

Abstract: In human and guinea-pig parturition, progesterone withdrawal and estrogen action are not mediated by changes in their circulating levels. Instead, these events might be promoted by changes in the responsiveness of the uterus and cervix to progesterone and estrogen via changes in their receptors. In this study, the guinea-pig model was used to investigate whether high levels of progesterone and estrogen at term are associated with regional changes in PR and ERalpha levels in uterus and cervix. PR and ERalpha pr… Show more

“…The uterine cervix undergoes dramatic adaptation processes during pregnancy, parturition and postpartum (Shi et al 2000, Montes et al 2002, Rodríguez et al 2003. There are striking changes in uterine vascular permeability, growth, density, vasodilatation and blood flow as a physiological response to cellular modifications, especially during pregnancy (Pavelock et al 2001, Reynolds et al 2002.…”

“…The feasibility of using commercial kits prepared for human plasma E 2 and P 4 determination in guinea pigs has previously been validated by others (Rodriguez et al 2003a;Rodriguez et al 2003b). The maximal interassay coefficient of variation for P 4 was 9.8% and for E 2 it was 7.5%.…”

“…For granulosa cell proliferation, we used an anti-Ki67 affinitypurified rabbit polyclonal antibody generated and tested in our laboratory (Varayoud et al 2008, Rivera et al 2011. The specificity of each antibody was tested using western blot analysis of protein extracts (Rodríguez et al 2003) obtained from intact uterine or gonad samples of ewes (data not shown). Each immunohistochemical run included positive tissues and negative controls replacing the primary antibody with nonimmune serum (Sigma-Aldrich).…”
