Collaborative study on rat sperm motion analysis using Cellsoft Series 4000 semen analyzer.

Abstract: A collaborative study was conducted to determine useful and sensitive rat sperm motion parameters in a CellSoft Series 4000 semen analyzer to detect the effects of compounds on sperm motion. The effects on the sperm motion parameters were investigated using alpha-chlorohydrin, boric acid, ethinylestradiol, ethyl methanesulfonate, nitrazepam, nitrobenzene, ornidazole, sulfasalazine or valproic acid which are well known to induce reproductive or testicular toxicities. All compounds used in this study decreased p… Show more

“…To elucidate the sperm motion parameters that are useful and the most sensitive indicators of the effects of various toxicological chemicals on sperm, collaborative studies in Japan were conducted using the CellSoft Series 4000 semen analyzer and the Hamilton-Thorne Sperm Analyzer (HTM-IVOS), which have nine sperm motion parameters each (Ban et al, 2001;Kato et al, 2001). The CASA system has allowed us to identify toxicant-induced changes in sperm motion that were previously undetected by manual sperm analysis.…”

“…Therefore, linearity may be a uniquely sensitive indicator of dog sperm motion. ALH max and ALH mean were slightly increased, although not to a significant extent, even though these parameters were decreased in rats by -chlorohydrin or other compounds (Ban et al, 2001). The cause of this different direction of change seems to originate from different shape of sperm between rats and dogs.…”

“…VCL or linearity were significantly decreased by -chlorohydrin treatment. In the above-mentioned collaborative study of rats, VCL was one of the most sensitive parameters, but linearity exhibited little or no change on treatment with 9 compounds including -chlorohydrin (Ban et al, 2001). Therefore, linearity may be a uniquely sensitive indicator of dog sperm motion.…”
