Shigella sonnei is a major agent of diarrhoeal disease in developed as well as in developing countries. Several phenotypic methods to define strain differences have been applied to this species of Shigella including, more recently, analysis of extrachromosomal and chromosomal DNA. In this study, 432 endemic and epidemic strains isolated between 1975 and 1991 in Italy, France and Switzerland were submitted to rRNA gene restriction pattern analysis, after digestion of whole-cell DNA by Hinc II, and to concomitant biotyping. Thirteen ribotypes, H1 to H13, and five biotypes, a, d, e, f, g, were detected. Ninety-five percent of the sporadic strains were assigned to ribotypes H1 to H4, which could be subtyped, except for H4, in different biotypes. Strains from each of seven different outbreaks had indistinguishable ribotype-biotype patterns. In contrast, 65 strains, isolated in Sicily in 1980 over an extended period of apparently epidemic increase of isolations and which had previously been considered to be a single bacterial clone on the basis of resistance pattern and phage type, were found to belong to two different and scarcely related ribotypes. Ribotyping and biochemical subtyping appear to be a useful epidemiological tool in studies on the circulation and distribution of strains of S. sonnei.

Shigella sonnei is a major cause of diarrheal disease in developed as well as in developing countries. Epidemiologic studies of this organism have been limited by the lack of a simple and effective method for comparing strains. In this study, we have compared different molecular typing methods, i.e., plasmid profile analysis, restriction endonuclease analysis of plasmids, rRNA gene restriction analysis (ribotyping), pulsedfield gel electrophoresis (PFGE), and enterobacterial repetitive intergenic consensus (ERIC) sequence-based PCR (ERIC-PCR) for typing 20 clinical isolates of S. sonnei collected from six incidents of infection. PFGE and ERIC-PCR fingerprintings had the highest discriminatory power for discrimination of epidemiologically related isolates from epidemiologically unrelated strains of S. sonnei, and both gave seven distinct strain types among these isolates and the type strain of the species. Plasmid study and ribotyping produced only six and two distinct patterns, respectively, among these strains. All of these molecular typing techniques demonstrated an identical fingerprint for eight temporally related sporadic isolates. It is possible that these temporally related isolates belonged to a single bacterial clone and circulated obscurely through the community. Our results indicate that the ERIC-PCR technique represents a rapid and simple means for typing S. sonnei with a level of discrimination equivalent to that of PFGE but greater than those of plasmid profile analysis, restriction endonuclease analysis of plasmids, and ribotyping.

Colicin typing, biotyping, phase typing and in vitro antimicrobial susceptibility testing were performed on clinical isolates of Shigella sonnei from three different geographical locations. Results of 93 strains from Kuwait, 61 strains from Saudi Arabia and 23 strains from Bangladesh were compared. Of 169 strains of S. sonnei tested, 75% belonged to colicin type 6/11. It was the most common colicin type in all three countries. However, differences in antimicrobial susceptibility patterns were observed. The resistance pattern in 62% of resistant strains from Kuwait was tetracycline – trimethoprim/sulpha-methoxazole – trimethoprim (TE-TMP/SMZ-TMP). Saudi Arabian strains were of the same resistance pattern with additional resistance to ampicillin (AM) AM-TE-TMP/SMZ-TMP (34%) which was seem more often than TE-TMP/SMZ-TMP (26%). Most of the strains (65%) from Bangladesh were sensitive to all the antimicrobial agents against which they were tested. Of the 8 strains which showed resistance, 50% were resistant to tetracycline alone. Study of carbohydrate fermentation of 12 sugars which were incubated for 21 days did not aid in discriminating different biotypes.
