Colchicine photosensitizes covalent tubulin dimerization

Abstract: Pure rat brain tubulin can be cross-linked by ultraviolet irradiation of tubulin-colchicine complexes at the high-wavelength maximum of colchicine to form covalent dimers greater than trimers greater than tetramers. With colchicine concentrations approximately 3 x 10(-4) M (mole ratio to tubulin 3-12) and irradiation for 5-10 min at 95-109 mW/cm2, the yield of dimers is 11-17% and of trimers is 4-6% of the total tubulin. The oligomers show polydispersity and anomalously high apparent molecular masses that conv… Show more

“…Whether these bands are precursor peptides or provide other binding surfaces remains to be determined. Moreover, the region of a-tubulin that is near enough to the colchicine-binding site on ,B-tubulin to become labeled under mild denaturing conditions (6,9,10) remains to be identified. Nonetheless, our findings identify two regions of 3-tubulin that are important structural elements of the colchicine-binding site.…”

“…These results suggested the possibility that the colchicine-binding domain on 3-tubulin might be near a-tubulin. That proposal was strengthened by the finding that colchicine photosensitized the formation of covalent tubulin dimers, trimers, and tetramers when the colchicine-tubulin complex was irradiated (10). Predominant labeling of (3-tubulin has also been demonstrated with chloroacetates of 2-or 3-demethylthiocolchicines (11).…”

“…Ice-water cooled samples were irradiated under 2.5 cm of 20% CuS04-5H20 (as far ultraviolet and infrared filter) for 5 min at 95-W lamp power. Stained 8% gels were quantified on a Beckman DU-8 gel scanner as described (Wolff et al" 1992).…”
