Coinfection withSchistosoma mansoniIs Associated with Decreased HIV-Specific Cytolysis and Increased IL-10 Production

Abstract: Impaired virus-specific immune responses have previously been observed with Schistosoma mansoni coinfection. We characterized Gag-specific responses in HIV-1-positive Ugandans with and without S. mansoni coinfection. We observed no significant difference in the frequency of IFN-γ CD8+ T cells between the two groups. Interestingly, expression of CD107, a marker for cytolytic activity, was significantly lower in volunteers with S. mansoni coinfection compared with those with HIV-1 infection alone (p = 0.002). In… Show more

“…Few studies have reported the relationship between cytokines in individuals with HIV and parasitic intestinal infections. Some authors evaluated cytokines and their relationship with parasites such as Schistosoma mansoni, Wuchereria bancrofti, and Trichinella spiralis (71)(72)(73)(74)(75)(76)(77)(78)(79). No correlation between serum INF-γ, IL-2, and IL-10 levels and intestinal parasitic infections was seen.…”

“…The detection of Gag-specific IL-10 production was performed using PBMC (1 ϫ 10 6 ) incubated with Gag peptide pools for 2 h at 37°C in 5% CO 2 in the presence of costimulatory anti-CD49d and anti-CD28 (1 g/ml; BD Biosciences) followed by GolgiStop (BD Pharmingen) for 12-14 h as previously described to be the optimal incubation period for the detection of IL-10 without affecting the viability of the cells (25). LPS (1 ng/ml; Sigma-Aldrich) and medium alone without Ag stimulation were used as positive and negative controls, respectively.…”

“…We have previously demonstrated that S. mansoni coinfection was associated with increased frequency of IL-10-positive HIV-specific CD8 ϩ T cell responses (25). Therefore, only individuals without serologic evidence of S. mansoni infection were evaluated in the current study to exclude schistosomiasis as a contributing factor in HIV-specific IL-10 production.…”

“…We first determined the frequency of Gag-specific IL-10-positive CD8 ϩ T cells in 61 ARV naive HIV-1-infected volunteers (25). Degranulation and IFN-␥ production by Gag-specific CD8 ϩ T cells were also determined concurrently.…”
