Cohort study of electroencephalography markers of amyloid-tau-neurodegeneration pathology

Abstract: Electroencephalography signatures of amyloid-β (Aβ), tau, and neurodegenerative pathologies would aid in screening for, tracking progression of, and critically, understanding the pathogenesis of dementia. We hypothesized that slowing of the alpha peak frequency, as a signature of hyperpolarization-activated cyclic nucleotide gated “pacemaker” channel activity, would correlate with amyloid and tau pathology burden measured by amyloid (Pittsburgh Compound B) and tau (MK-6240) PET or CSF biomarkers. We also hypot… Show more

“…Their results revealed that theta activity in fronto-central regions was higher in tau/neuroinflammation-positive patients. Consistent findings across studies also highlight that both β-amyloid and tau/neuroinflammation were associated with slowing of brain rhythm activity in aging (Tanabe et al, 2020). Here we replicated the association for theta daily fluctuations only with tau/neuroinflammation, which would indicate that tau is associated with different features of theta brain rhythms.…”

“…An a priori sample size was not conducted, as these data were collected as part of a study that also collected resting state EEG that is already published. 14 All available data from the cohort study were included in this work.…”

“…Similar network alternations associated with biofluid markers have been put forward. For example, increased CSF p- and t-tau levels were correlated with EEG slowing and decreasing synchronization in patients with cognitive impairment ( Smailovic et al, 2018 ; Tanabe et al, 2020 ); the combined p-tau/Aβ42 ratio exhibited a stronger correlation with the slow-frequency band in elderly individuals ( Stomrud et al, 2010 ). Beyond plasma t-tau, we also found some specific correlations between other plasma biomarkers (NfL and α-syn) and EEG parameters in individual band power.…”
