Cohort Profile: The Lothian Birth Cohorts of 1921 and 1936

Abstract: This cohort profile describes the origins, tracing, recruitment, testing and follow-up of the University of Edinburgh-based Lothian Birth Cohorts of 1921 (LBC1921; N = 550) and 1936 (LBC1936; N = 1091). The participants undertook a general intelligence test at age 11 years and were recruited for these cohorts at mean ages of 79 (LBC1921) and 70 (LBC1936). The LBC1921 have been examined at mean ages of 79, 83, 87 and 90 years. The LBC1936 have been examined at mean ages of 70 and 73 years, and are being seen at… Show more

“…Validation of age-associated CpGs was done in two independent longitudinal cohorts; the Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) [18] and the Lothian Birth Cohort (LBC) [19]. The PIVUS cohort measured methylation data from 196 individuals at age 70 and 80, and the LBC cohort, including two sub-cohorts of 906 and 436 individuals at baseline, measured blood samples at three time points, with mean ages of 70 and 79 at baseline (Table S1).…”

“…This study reports analysis of data collected from members of the Lothian Birth Cohort 1936 (LBC1936; [Deary et al, 2012, 2007]), a longitudinal study of ageing in the Edinburgh and Lothians area of Scotland, UK. Written informed consent was obtained from all participants, and the overall study was approved by the Lothian Research Ethics Committee (LREC/2003/2/29) and the Multi‐Centre Research Ethics Committee for Scotland (MREC/01/0/56).…”

“…Our Discovery cohort consisted of genome-wide association study (GWAS) data relating to 6,732 (after QC) cognitively healthy subjects participating in the "Generation Scotland: Scottish Family Health Study" (GS:SFHS) 24 . The Replication cohort consisted of independent GWAS data relating to 1,003 (after QC) cognitively healthy subjects participating in the Lothian Birth Cohort 1936 (LBC1936) 25 . Mean age at assessment was 55 years (standard deviation (SD)=11.35) in GS:SFHS and 69.6 years (SD=0.8) in LBC1936.…”
