“…Reports of disease-associated alleles are not consistent, but associations appear to be strongest with alleles in the HLA-DR and -DQ loci. This has also been suggested by studies in transgenic mouse [30,52,85-87]. …”

“…A role for HLA-DQ polymorphism was shown with human thyroglobulin-induced EAT in HLA-DQ*0301/DQB1*0302 (DQ8), but not HLA-DQ*0103/DQB1*0601 (DQ6), transgenic mice [52]. In summary, DR3 and DQ8 alleles are found to be susceptible, whereas DR2, DR4 and DQ6 alleles are resistant [30,87]. Studies on EAT-developing mice showed the differential effects of class II molecules on EAT induction.…”

“…Similarly, we have shown that the presence of an HLA-DQ8 transgene plays a moderating role in HLA-DR3-mediated response to hTg in DR3/DQ8 double transgenic mice [47]. The existence of Tregs influencing EAT induction in the context of this H2A b class II molecule [23] suggested that they may be responsible for the suppressive effect of H2A expression on H2E-mediated thyroiditis.…”

“…In Her-2 tolerant, EAT-resistant C57BL/6 mice also harboring the HLA-DRB1*03:01 transgene to encode EAT susceptibility (15), the (H2 b -Her-2xDR3)F 1 mice withstood tumor challenge again only after Treg depletion and Her-2 DNA vaccination (16). Although the presence of the DR3 allele in F 1 mice increased the autoimmune response, it was moderated by the EAT-resistant A b allele, consistent with EAT-resistant DQ8 (HLA-DQA1*03:01/DQB1*03:02) transgene downregulation of DR3-mediated thyroiditis (17). In all three combined models, immune stimuli from concurrent tumor regression and EAT development had a detectable, mutually enhancing effect.…”
