Cocaine Increases the Intracellular Calcium Concentration in Brain Independently of Its Cerebrovascular Effects

Abstract: Cocaine abuse increases the risk of life-threatening neurological complications such as strokes and seizures. Although the vasoconstricting properties of cocaine underlie its cerebrovascular effects, the mechanisms underlying its neurotoxicity remain incompletely understood. ] i in the brain. The effects of cocaine were compared with those of methylphenidate, which has similar catecholaminergic effects as cocaine (except for serotonin increases) but no local anesthetic properties, and of lidocaine, which has s… Show more

“…To assess cellular oscillations, we measured LFOs of Ca 2+ i fluorescence using the calcium indicator Rhod 2 -AM locally microinjected into the somatosensory cortex (27). The LFO power spectra of CBFv, HbO 2 , and HbR within the Rhod 2 -loaded tissue area were almost identical to those in surrounding areas without Rhod 2 loading (SI Appendix, Fig.…”

“…1E and Fig. 5, 6 of the 10 rats received microinfusion of the Ca 2+ i indicator Rhod 2 -AM (100μM, 3 μL/min; Molecular Probes) locally in the cortex (27) before the dura was removed, whereas other the 4 rats were used to assess effect of PCO 2 on vascular dilation and to measure local field potentials (LFP) for assessing changes in neuronal activity from normocapnia to hypercapnia. Hemodynamic parameters were compared between an ROI near the Rhod 2 injection spot (red circle and red curves) and a distant ROI (light blue circle and light blue curves) as shown in SI Appendix, Fig.…”

“…It causes vasoconstriction due to its potent sympathomimetic properties. It prevents the reuptake of noradrenaline, serotonin, and dopamine at presynaptic nerve terminals and increases the release of calcium from the sarcoplasmic reticulum in cerebral vascular smooth muscle cells, resulting in vasoconstriction [5,6]. Endothelin-1 is another important mediator of cocaine-induced cerebral vasospasm [7].…”

“…Effects may be transient or permanent and occur as a result of decreases in mean arterial blood pressure, glucose utilization, blood volume, tissue oxygenation, and cerebral blood flow due to vasoconstriction in addition to an independent increase in intracellular calcium concentration associated with cell death. 9 Conversely, therapeutic hypothermia has long been believed to be capable of reducing primary brain injury and preventing secondary inury, 10 and its use even in stroke patients is an area of ongoing study and scientific interest without definitive conclusions. 11 Therapeutic hypothermia is theorized to combat global ischemia in several unique ways.…”
