Cobalt‐Catalyzed Intermolecular Oxidative Isocyanide Insertion with Two Amines: An Approach to Guanidines

Abstract: A novel and efficient cobalt-catalyzed oxidative isocyanide insertion with amines via two C À N bond formation reactions under ultrasound irradiation conditions has been developed. This protocol provides a simple, clean and general way to synthesize guanidines or their corresponding hydrochlorides from simple starting materials under ultrasonic conditions. This chemistry could also be applied to an efficient synthesis of optically active guanidines without loss of enantioselectivity.

“…An interesting example is the synthesis of unsymmetrically substituted arylguanidines 90 using ac obalt catalyst under ultrasound heating (Scheme 23). [52] Thep rocess involves the imidoylative coupling of an aliphatic and an aromatic amine using (t-BuO) 2 as the oxidant (Scheme 24). Thei nitially formed aromatic C-amidinoylcobalt intermediate 92 can be formed by either aT ype IoraTy pe II pathway.…”

“…The combination of a perester and catalytic Co(II) promotes reaction of isocyanates and a pair of amines, one of which is always an aniline, to generate guanidines . The described mechanism involves addition of the aniline to the carbon of a Co(II) isocyanate, resulting in an amidine.…”
