Coated microneedles for transdermal delivery

Abstract: Coated microneedles have been shown to deliver proteins and DNA into the skin in a minimally invasive manner. However, detailed studies examining coating methods and their breadth of applicability are lacking. This study's goal was to develop a simple, versatile and controlled microneedle coating process to make uniform coatings on microneedles and establish the breadth of molecules and particles that can be coated onto microneedles. First, microneedles were fabricated from stainless steel sheets as single mic… Show more

“…Using methods described in detail previously [4], microneedles were cut into 75 μm thick stainless steel sheets (Trinity Brand Industries, SS 304; McMaster-Carr, Atlanta, GA, USA) using an infrared laser (Resonetics Maestro, Nashua, NH, USA). Holes through microneedle shafts to form "pockets" were similarly cut.…”

“…Single microneedles, and arrays of multiple microneedles, were coated using a micron-scale dip-coating process developed previously [4]. Coatings were formed by dipping the microneedle into a coating solution once, unless otherwise noted.…”

“…But, it has a number of disadvantages associated with it. Hypodermic needles often cause pain, transmit pathogens through needle re-use and require medical expertise to use (Gill and Prausnitz, 2007). Transdermal drug delivery is both painless and user-friendly.…”

“…Many microneedle systems (e.g. coated 41 , dissolvable 42 , swellable 43 ) incorporate the drug into the device in a solid-state, extending the shelf-life compared to drug solutions in syringes 44 . Post-insertion, these needles are no longer sharp 42,43 , thus preventing reuse, needle-stick injury as well as reducing waste management issues.…”
