Coagulansin-A has beneficial effects on the development of bovine embryos in vitro via HSP70 induction

Abstract: SynopsisCoagulansin-A (withanolide) is the steroidal lactone obtained from Withania coagulans which belong to Solanaceae family. The present study investigated the effects of coagulansin-A on bovine oocyte maturation and embryo development in vitro. All these oocytes were aspirated from the ovaries obtained from Korean Hanwoo cows at a local abattoir. To determine whether coagulansin-A has beneficial effects on bovine oocyte maturation in vitro, 355 oocytes per group (control and treated) in seven replicates w… Show more

“…The numbers of apoptotic and total cells are key factors for embryo quality; the higher the number of cells in an implanted embryo, the higher the chance of live offspring being delivered (van Soom et al 2007). In the present study, coa-A treatment in IVM media at a heat stress of 40.5˚C for 20 h improved blastocyst quality by increasing the total number of cells and decreasing the apoptotic index compared with the control group, which is in agreement with our previous findings at a normal culture temperature (Khan et al 2016).…”

“…The goal of this study was to determine the protective effect of coa-A against heat stress in IVM media for blastocyst development. For this purpose, the optimal concentration of coa-A (5 μM) was selected based on our previous data (Khan et al 2016). Oocytes treated with 5 μM coa-A were compared with the untreated control group.…”

“…This result is consistent with a recently published study that addition of coagulansin‐A, a ROS‐scavenging compound, to IVM media induced HSP70 expression in bovine embryos, and this protein after inducing in the culture system improved the embryo quality and efficiency (Khan et al . ).…”
