CO2 hydration by immobilized carbonic anhydrase

Abstract: The enzyme carbonic anhydrase (isoform II) from bovine and human erythrocytes was immobilized using different covalent coupling methods on inert matrices. Immobilized carbonic anhydrase may enable concentration of CO2 for Rubisco (ribulose-1,5-bisphosphate carboxylase/oxygenase)-catalysed fixation in bioreactors. In the present study the activity of carbonic anhydrase with respect to hydration of CO2 using soluble and immobilized enzymes was determined. The stability of the immobilization matrix, the propertie… Show more

“…Immobilised PEG‐bound ATP derivatives, as well as enzymes for ATP regeneration, were entrapped in ultrafiltration membranes, resulting in model reactions in a turnover number of 19 500 mol/mol 93. An approach combining electrochemical potential energy generated by solar cells with immobilised F o F 1 –ATPase complex on semipermeable membranes yielded enzymes that retained up to 90 % of activity and good stability under process conditions 96…”

“…Aside from cost, two challenges often encountered by immobilization technologies are the loss of enzyme from the immobilization matrix over time, 214 called "enzyme leaching," and underestimating or discounting enzyme activity losses during the immobilization process. Immobilization should demonstrate orders of magnitude longevity improvement to justify the additional processing and cost.…”

“…In parallel, to further optimize the process and to improve storage, thermal and/or pH stability of the enzyme, immobilization on supports is of great interest . TK immobilization has been investigated on organic or semiorganic supports, such as activated sepharose, Eupergit‐C, Amberlite XAD‐7 and Nylon, and polyacrylamide gel . We have also reported the immobilization of TK yst and TK eco on an inorganic support, layered double hydroxides (LDHs).…”
