Co-localized confocal Raman spectroscopy and optical coherence tomography (CRS-OCT) for depth-resolved analyte detection in tissue

Abstract: Abstract:We report the development of a combined confocal Raman spectroscopy (CRS) and optical coherence tomography (OCT) instrument (CRS-OCT) capable of measuring analytes in targeted biological tissues with sub-100-micron spatial resolution. The OCT subsystem was used to measure depth-resolved tissue morphology and guide the acquisition of chemically-specific Raman spectra. To demonstrate its utility, the instrument was used to accurately measure depth-resolved, physiologicallyrelevant concentrations of Teno… Show more

“…A custom built CRS-OCT (confocal Raman spectroscopy and optical coherence tomography) instrument integrated with a white-light camera was used to acquire data in this study. 19 The axial and lateral resolutions of the CRS subsystem are 19 and 3 μm, respectively. The OCT subsystem has an axial resolution of 8 μm and a lateral resolution of 40 μm, and the white-light camera offers a lateral resolution of 2 μm.…”

“…A custom Matlab script was used to process raw spectral data, as outlined previously. 19 The contributions of tenofovir to each Raman spectrum were established by ordinary least squares fitting. A representative fit with the Raman spectral contributions of Tenofovir, gel, and vaginal fluid simulant is displayed in Figure 2.…”

“…Various illumination and detection schemes have been combined with RS providing different trade‐offs in resolution, penetration depth, and contrast. Confocal detection, for example, provides high spatial resolution up to approximately one transport length below the sample surface . When measuring bone, confocal RS is primarily sensitive to the cortex, as the transport length of near‐infrared light in bone is only about 500 microns .…”

“…It is highly desirable to combine the RS and OCT techniques for simultaneously acquiring both tissue morphology and biochemical information on the same tissue in real‐time. Previous studies on the combined RS and OCT are largely limited to the bench‐top optics , and the 2 optical techniques are conducted sequentially with lengthy acquisition time , unsuited for rapid in vivo tissue measurements in clinical settings. Since the biochemical and morphological profiles of distinctive anatomical regions in the oral cavity can be highly functionally specialized and exhibit significant variations in architectural properties and cell types (eg, tissue thickness, distinct epithelium types, vascularity, papillae, bone, etc.)…”
