Co-expression of somatolactin mRNAs in the pituitary gland of gilthead sea bream

Abstract: The expression of mRNAs of the two types of somatolactin which have been found, up to the present, in the pituitary of gilthead sea bream (Sparus aurata) have been analyzed by in situ hybridization (ISH). The method of non-radioactive ISH, which we optimize in this study, uses oligonucleotides labelled in the 5' end with biotin or dioxygenin as probes. This allows simple and double ISH of high specificity and sensitivity to be performed. The distinct somatolactin oligoprobes used present a strong signal fundam… Show more

“…Nevertheless, a distinct differential secretion of GH could be suggested by the differences among the hypophysial sizes/volumes and the number of adenohypophysial cells. Therefore, no asynchronic growth-related changes were observed in the distribution of GH cells, nor in the distribution of lactotropes (Herrero-Turrión et al 2003a) and SL cells (Herrero-Turrión et al 2002). Therefore, although GH is essential for growth and development throughout the development of fish, in gilthead sea bream the asynchronous growth cannot be due to different distributions of lactotroph cells.…”

“…Hybridization procedures were as described elsewhere (Herrero-Turrión et al 2002, 2003a. Before carrying out the hybridization, the sections were postfixed with PFA and acetylated in a solution with 1.5% triethanolamine, 150 mM NaCl and 0.25% acetic anhydride to avoid false-positive results.…”

“…Adicionalmente, uma nova variante de somatolactina foi descoberta em adultos de Sparus aurata (Cavari et al (2000), a qual difere em 11 aminoácidos da primeira forma isolada, e ambas são coexpressas nas mesmas células somatolactínicas (Herrero-Turrión et al, 2002).…”
