Co-delivery of siRNA and therapeutic agents using nanocarriers to overcome cancer resistance

Abstract: Summary There are two main mechanisms by which cells become multidrug resistant (MDR): by increasing drug efflux pumps on the cell membrane and by increasing anti-apoptotic pathways. The use of nanotechnology to develop nanodelivery systems has allowed researchers to overcome limitations of antineoplastic drugs by increasing the solubility of the drug and decreasing the toxicity to healthy tissues. By encapsulating drugs into nanoparticles that bypass the efflux pumps, drug efflux is reduced, hence increasing … Show more

“…Attempts to develop nanocarriers for co-delivery of small interfering RNA (siRNA) and small molecule drugs continue to grow, particularly in recent years [185]. Combined gene and chemotherapy requires nanocarriers to be inert in nature, non-immunogenic, non-toxic, capable of effectively condensing and immobilizing nucleic acids, and encapsulating small molecules [186].…”

“…130,150,151 As there will be reviewed, the delivery of multiple therapeutic species with a single device share some of the ideas developed for gated systems. Hence, as the incorporation of therapeutic nucleic acids onto MSNs is mostly based on the electrostatic multilayer deposition of NAs onto MSNs, it could be foreseen a parallel development for the systems that combine cytotoxic delivery plus gene modulation.…”
