Co-circulation of all the four dengue virus serotypes and detection of a novel clade of DENV-4 (genotype I) virus in Pune, India during 2016 season

Abstract: Dengue is the most common mosquito-borne viral infection in tropical and sub-tropical countries. In recent years, India has reported increased incidences of concurrent infection with multiple serotypes of dengue viruses (DENV). In the present study, we have characterized DENV circulating during a single season of 2016 in Pune, India. A total of 64 serum samples from NS1 ELISA positive dengue patients were used for PCR amplification of CprM region of the viral genome and sequencing. Phylogenetic analysis docume… Show more

“…We used CprM region–based PCR for the detection of viral RNA . Two earlier studies from India, including ours, and one from Mexico used this PCR for identifying viremia among serologically diagnosed patients with dengue. DENV RNA positivity was found to be 48.6% (53/109)and 52% (71/136) among NS1 ELISA positives or 59.5% among 117 NS1 and 4 IgM positives (immunochromatographic rapid test) .…”

“…The study was conducted during two dengue seasons (2016 and 2017). All four DENV serotypes were circulating during both the seasons (unpublished observations). After informed consent, leftover serum samples from voluntary blood donors were obtained from the blood bank of Bharati Vidyapeeth Hospital (Pune, India) and stored at ‐70°C in aliquots.…”

“…All DENV-4 genotypes sequenced were found to be genotype I with the earliest described from Tamil Nadu (Vellore, 1961). (Shrivastava et al, 2018) Thereafter this genotype has been described from all across India. However, Andhra Pradesh had a genotype II sequence reported in 2014.…”

“…There are four dengue viruses, denoted as DENV-1, DENV-2, DENV-3, and DENV-4 [5]. The genome structure of DENV has positive sense RNA genome of 10.6 to 11.0 kb [6].…”
