Co-circulation in a single biome of the Juquitiba and Araraquara hantavirus detected in human sera in a sub-tropical region of Brazil

Abstract: Hantaviruses is an emerging infectious disease. Although HCPS has been reported in several regions of Brazil, more cases of HCPS have recently been reported in Minas Gerais than in any other state. In 2009, we analyzed 27 samples presenting antibodies against hantaviruses. These samples originated from 688 symptomatic patients, as determined based on the Hemorrhagic Fever Protocol. A subsequent SYBR Green-based real-time RT-PCR demonstrated the presence of the virus in 22 of the samples. Among the RT-PCR-posit… Show more

“…Among many hantavirus genotypes, those with disease association as determined by virus genome demonstration in patient samples were identified by published literatures [Torrez‐Martinez et al, ; Rivers et al, ; Nelson et al, ; Knust and Rollin, ; Figueiredo et al, ; Atkinson et al, ; de Araujo et al, ; Guterres et al, ; Limongi et al, ; Oliveira et al, ; Vial et al, ]. Based on which, 13 genotypes were selected for further analysis.…”

“…To our knowledge, no cases of HPS have been associated with Jabora virus (JABV), harbored by Akodon montensis (tribe Akodontini). Akodon montensis is widely distributed in the IAF, a vast neotropical biome that extends into eastern Paraguay, and into the temperate grass and shrubland biomes of Argentina [ 16 , 17 ]. However, another member of the Akodontini ( Necromys lasiurus ) has been associated with HPS cases associated with Araraquara virus in Brazil [ 18 ].…”

“…In the moist broadleaf forest biomes from Venezuela and Colombia into Paraguay, distinct rodent genera harbor JABV, Maporal or Necocli hantaviruses [ 12 , 23 25 ]. In the IAF and coastal Atlantic Forest in Brazil, several phylogenetically distinct lineages of RBO-HV, (e.g., JUQV, Oran and Lechiguanas hantaviruses), have been identified in a variety of Oligoryzomys species [ 16 , 17 ]. In the Reserva Natural del Bosque Mbaracayú (RNBM) in eastern Paraguay, the reservoirs of JABV and JUQV, Akodon montensis and Oligoryzomys nigripes respectively, are sympatric [ 12 ].…”

“…Currently, orthohantavirus commercial serological kits are based on nucleoprotein detection however the cross-reactivity among hosts infected by HCPS and/or HFRS-associated orthohantavirus could not be discarded. In our study, we experimentally immunized mice with recombinant nucleoprotein from Juquitiba genotype, a HCPS-associated orthohantavirus that mainly circulates in Brazil [39, 40], eliciting a polyclonal response able to generate antibodies that recognized JHNP and SHNP ( Fig 2 ). Based on this cross reaction and considering that Juquitiba genotype nucleoprotein is highly conserved among ANDV and SNV (95% and 86% of similarity, respectively), both orthohantavirus associated to HCPS in Americas, we believe that Juquitiba genotype could be used as HCPS model.…”
