Clusterin/Apolipoprotein J immunolocalization on carotid artery is affected by TNF-alpha, cigarette smoking and anti-platelet treatment

Abstract: BackgroundClusterin (CLU) /Apolipoprotein J is a protein biosensor of oxidative stress and inflammation, which is upregulated in many pathological processes including atherosclerosis. Previous studies have shown that in aortic tissue, CLU expression increases with atherosclerotic lesion progression and it has been coupled with vascular damage and coronary artery disease. A few studies enter into CLU and carotid atherosclerosis while the apolipoprotein’s expression on human carotid tissue and its association wi… Show more

“…Apo J has been reported to be closely related to cardiovascular diseases, such as atherosclerosis and restenosis after angioplasty. 14 , 15 Ishikawa et al 7 revealed the distribution of Apo J in the extracellular matrix of endarterium in human atherosclerotic aorta, and its potential protective role against human atherosclerosis by cholesterol transport from the aortic wall to the liver. It has been reported that ApoJ is increased in tissue injury and cell stress, and plays vital role in protection against oxidative stress, cell lysis and apoptotic cell death.…”

“…Venous serum blood samples were collected after overnight fasting and were stored frozen in aliquots at -80°C until further processing. Sensitive ELISA sandwich immunoassay (Immundiagnostik AG, Bensheim, Germany) was used for determining TNF- α according to manufacturer's instructions [ 31 ].…”

“…A proteomic analysis has found no changes in the amount of exchangeable apolipoproteins in the plasma lipoproteins from patients with carotid artery atherosclerosis [40]. In human carotid plaques, several apolipoproteins, are expressed including apo AI [47] and apoJ [51]. ApoJ and apoE are released by carotid plaques in a higher degree relative to their expression in non-atherosclerotic arteries [49].…”
