Clues on the coding of reward cues by the nucleus accumbens

Abstract: The study by Calipari et al. in PNAS provides a glimpse into the in vivo pattern of neuronal activation in the mouse nucleus accumbens during the acquisition and expression of learned reward-context associations (1). The nucleus accumbens sits in the ventral part of the mouse striatum, and is required for reward-motivated learning. The activity of neurons in this region is thought to code information on the reward value of cues and contexts. Two distinct subpopulations of striatal neurons can be identified bas… Show more

“…The dopamine receptors D1r and D2r exert opposite effects on neuronal excitability with D1r increasing and D2r decreasing it (Alvarez, 2016). D1r and D2r expressing neurons in the striatum also have distinct expression patterns for endogenous opioids; enkephalin is mainly co-expressed in D2r whereas dynorphin is mainly co-expressed in D1r-expressing neurons (Surmeier et al, 1998;Gerfen and Surmeier, 2011;Gangarossa et al, 2013).…”

“…In the experimental paradigm, mice were exposed to the saline and cocaine chambers separately since the 2-separated chamber apparatus was used during dopamine measurements. However, in many previous studies, a 2- or 3-connected chamber apparatus, where mice were allowed to freely explore the entire apparatus, was used for the assessment of CPP expression ( Prus et al, 2009 ; Alvarez, 2016 ). The difference in the CPP apparatus between the 2-separated and 2-connected chambers could affect the dopamine response in the NAc.…”

“…Thus, photometry signals in vivo are most likely a reflection of 1–3 action potentials and the signal detection is biased toward synchronized events that can summate in time. Photometry signals might reflect the activity of ensembles rather than single neurons (Alvarez, 2016). While important to consider these factors, neither of these considerations undermines the main findings that co-activation of iSPNs and dSPNs is seen during a wide range of behaviors.…”
