Cloning of an acidic laccase gene (clac2) from Coprinus congregatus and its expression by external pH

Abstract: Coprinus congregatus synthesized and secreted high amounts of laccase under acidic culture condition (pH 4.1) transiently. We have cloned a genomic DNA fragment between the copper binding regions I and II of a laccase by PCR from C. congregatus. This fragment was used as a probe to clone a laccase cDNA from the acidic culture. The cDNA (clac2) consisted of 1817 bp which contains 1086 bp encoding 362 amino acids. The N-terminal sequence of the purified CLAC2 revealed that the first 16 amino acids were removed b… Show more

“…The pH 5.0~6.0 in the culture supernatant grown at 100 rpm was lower than pH 7.1~7.5 grown at 150 rpm. Many fungal laccases were induced and showed the higher activity at acidic conditions (Kim et al, 2001), and this result also followed the similar pattern. The higher mycelial concentration in the inoculum seemed to increase the laccase activity faster than the lower mycelial concentration at both 100 and 150 rpm of shaking conditions, but the differences were not significant (data not shown).…”

“…Chromosomal DNA from CL1407b was isolated [2] and digested with the Eco RV restriction enzyme, which has no recognition site in plasmid pBARGEM7‐1. The digested DNA fragments were separated on a 1% agarose gel, and DNAs longer than 4.5 kb were eluted from the gel.…”

“…Coprinellus congregatus is a mushroom‐forming basidiomycete, and its development is very easily manipulated in lab conditions [1]. There are two or more laccase isozymes implicated in the mushroom development, and the cDNA of the acidic laccase gene ( clac2 ) has been cloned [2]. Genetic transformation to phosphinothricin resistance has been carried out by restriction enzyme‐mediated integration (REMI) in C. congregatus , and several mutants that showed different growing patterns have been reported [3].…”

“…이 효소의 cDNA (lac2)를 확보하고 northern blot 분석 을 통하여 중성에서는 발현되지 않고 산성 조건에서만 발현됨 을 확인하였다 (Kim et al, 2001). 이 유전자의 발현 양상을 분 석하고자 promoter 및 genomic DNA 유전자를 cloning하였다 (Kim et al, 2006).…”
