Cloning of a Third Mammalian Na+-Ca2+ Exchanger, NCX3

Abstract: NCX3 is the third isoform of a mammalian Na ؉ -Ca 2؉ exchanger to be cloned. NCX3 was identified from rat brain cDNA by polymerase chain reaction (PCR) using degenerate primers derived from the sequences of two conserved regions of NCX1 and NCX2. The NCX3 PCR product was used to isolate two overlapping clones totalling 4.8 kilobases (kb) from a rat brain cDNA library. The overlapping clones were sequenced and joined at a unique Bsp106I restriction enzyme site to form a fulllength cDNA clone. The NCX3 cDNA clon… Show more

“…15,16 Three different ncx1, ncx2, and ncx3 genes have been identified in mammals. [17][18][19] Although the expression of the different NCX mRNAs in oligodendrocytes has been described, 20 their role during oligodendrocyte development has never been investigated. In the present study, we asked whether the three NCX isoforms, by modulating Na þ and Ca 2 þ homeostasis, might be involved in OPC maturation and myelin formation. To address these questions, we first examined their expression and, then, by using a patch clamp in whole-cell configuration and single-cell FURA-2 microfluorimetry, we studied their functional activity during oligodendrocyte and astrocyte development either in the human oligodendrocyte progenitor MO3.13 cell line or in primary rat OPC cultures.…”

“…3 Three NCX genes have been identified, NCX1, 4 NCX2 5 and NCX3. 6 In excitotoxic cell death, an increase in intracellular free calcium concentration ([Ca 2 þ ] i ) may directly cause activation of Ca 2 þ -dependent cysteine proteases, the calpains. Calpains modulate a variety of physiological processes, 7 and are important mediators of cell death.…”

“…Thus, transport of Ca 2+ out of the cell by Na + /Ca 2+ exchange generates inward current. The mammalian nervous system amply expresses three NCX isoforms (Nicoll et al 1990(Nicoll et al , 1996Li et al 1994), and therefore Na + /Ca 2+ exchange currents are expected whenever cytoplasmic Ca 2+ is elevated, including in response to activation of Group I mGlu receptors.…”
