Cytokinesis is the process that physically separates two sibling cells, ensuring the proper partitioning of the nuclear and cytoplasmic contents at the end of cell division. Cytokinesis requires a fine-tuned molecular machinery that has to be assembled with high spatiotemporal precision. Drosophila melanogaster is an ideal model system to investigate this cellular process. Genetic screens performed in spermatocytes, neuroblasts, and Schneider cells revealed numerous evolutionary conserved components. These genetically amenable systems have proven to be very useful to further elucidate the cellular and molecular mechanism of cytokinesis, significantly contributing to our current understanding of this important cellular process. As in other organisms, cytokinesis is largely dependent on the mitotic spindle, providing positional cues for cleavage furrow placement and progression. However, spindle-independent mechanisms could also be important during special cases of cytokinesis, such as asymmetric cell division. Thus, powerful fly genetics combined with single-cell analysis, live imaging, and biochemical assays will continue to provide important insights into the mechanism of cytokinesis. International audienceEukaryotic cells must possess mechanisms for condensing and decondensing chromatin. Chromatin condensation is particularly evident during mitosis and cell death induced by apoptosis, whereas chromatin decondensation is necessary for replication, repair, recombination and transcription. Histones are among the numerous DNA-binding proteins that control the level of DNA condensation, and post-translational modification of histone tails plays a critical role in the dynamic condensation/decondensation that occurs during the cell cycle. Phosphorylation of Ser10 in the tails of histone H3 has been extensively studied in many organisms. Interestingly, this modification is involved in both transcription and cell division, two events requiring opposite alterations in the degree of chromatin compaction. How does one and the same modification of histone H3 fulfil such roles? For instance, in interphase, phosphorylation of H3 correlates with chromatin relaxation and gene expression, whereas in mitosis it correlates with chromosome condensation. What is the kinase and under what circumstances does Ser10 becomes phosphorylated? Most importantly, what are the consequences of phosphorylation of this residue

The centrosome, also known as the microtubule organizing center of the cell, is a membrane-less organelle composed of a pair of barrel-shaped centrioles surrounded by electron-dense pericentriolar material. The centrosome progresses through the centrosome cycle in step with the cell cycle such that centrosomes are duplicated in time to serve as the spindle poles during mitosis and that each resultant daughter cell contains a single centrosome. Regulation of the centrosome cycle with relation to the cell cycle is an essential process to maintain the ratio of one centrosome per new daughter cell. Numerous mitosis-specific kinases have been implicated in this regulation, and phosphorlyation plays an important role in coordinating the centrosome and cell cycles. Centrosome amplification can occur when the cycles are uncoupled, and this amplification is associated with cancer and with an increase in the levels of chromosomal instability. The aurora kinases A, B, and C are serine/threonine kinases that are active during mitosis. Aurora A is associated with centrosomes, being localized at the centrosome just prior to the onset of mitosis and for the duration of mitosis. Overexpression of aurora A leads to centrosome amplification and cellular transformation. The activity of aurora A is regulated by phosphorlyation and proteasomal degradation.
