Cloning, Mapping, and Expression of ial, a Novel Drosophila Member of the Ipl1/aurora Mitotic Control Kinase Family

Abstract: The members of the Ipl1-aurora like kinase (IARK) subfamily are conserved serine/threonine kinases that play a key role in the control of chromosome segregation, centrosome separation, and cytokinesis from yeast to mammals. We report on the isolation of a new Drosophila member of the family, designated Ipl1-aurora-like kinase (ial) Phylogenetic analysis of kinase domains established that ial is more divergent from known mammalian IARKs than is aurora. Mapping based on examination of chromosomal aberrations, to… Show more

“…The conserved CPC, consisting of Aurora B kinase [Reich et al, 1999;Giet and Glover, 2001], Borealin-related (Borr) [Eggert et al, 2004;Hanson et al, 2005], Australin (Aust) [Gao et al, 2008], Inner centromere proteins (Incenp); [Chang et al, 2006] and Survivin (also called Deterin (Det)) [Szafer-Glusman et al, 2011], is localized to inner centromeres in metaphase, translocate to the spindle midzone and the equatorial cell cortex before moving to the presumptive site of cleavage furrow formation following metaphase-anaphase transition [reviewed in Ruchaud et al, 2007]. In C. elegans and mammalian cells, it was shown that the CPC component Aurora B phosphorylates the centralspindlin components Pav and Rac-GAP50C [Guse et al, 2005;Neef et al, 2006;Douglas et al, 2010].…”

“…Likewise in Schizosaccharomyces pombe, the single aurora kinase (Ark1) is associated with Ser10 phosphorylation in mitosis (Petersen et al, 2001). Multicellular organisms such as Caenorhabditis elegans and Drosophila melanogaster possess two aurora kinases -aurora A and aurora B (Schumacher et al, 1998a;Schumacher et al, 1998b;Glover et al, 1995;Reich et al, 1999), whereas mammals possess an additional spermatogenesis-dedicated kinase, aurora C (Giet and Prigent, 1999;Adams et al, 2001a). Examination of Ser10 phosphorylation with antiphosphoSer10 antibodies in nematode and fruit fly cells in which expression of aurora A or aurora B has been eliminated by RNA interference has revealed that aurora B but not aurora A is required for Ser10 phosphorylation Giet and Glover, 2001;Adams et al, 2001b).…”

“…The monopolar spindles arise as a result of failure of centrosome separation, which should occur in early mitosis [Glover et al, 1995]. Drosophila also expresses another aurora-related gene called Ipl1 aurora-like kinase, which is believed to play a role in cytokinesis [Reich et al, 1999]. The genome of Schizosaccharomyces pombe encodes one aurora gene, named aurora-related kinase-1 (Ark-1).…”
