Cloning Human Enamelin cDNA, Chromosomal Localization, and Analysis of Expression during Tooth Development

Abstract: Enamelin is the largest protein in the enamel matrix of developing teeth. In the pig, enamelin is secreted as 186-kDa phosphorylated glycoprotein, which is rapidly processed by enamel proteinases into smaller cleavage products. During the secretory stage of enamel formation, enamelin is found among the crystallites in the rod and interrod enamel and comprises roughly 5% of total matrix protein. Although the function of enamelin is unknown, it is thought to participate in enamel crystal nucleation and extension… Show more

“…1D). These results correlate well with previous studies of enamelin expression by in situ hybridization (9,56). The only difference is that maturation stage ameloblasts are positive for X-gal staining but negative for in situ hybridization.…”

“…There are three major secretory stage enamel proteins: amelogenin (5,6), ameloblastin (7), and enamelin (8,9). All belong to the secretory calcium-binding phosphoprotein (SCPP) gene family (10), and all are critical for proper dental enamel formation.…”

“…The translation initiation site has not been determined experimentally for ENAM, but we found an authentic SP sequence of 16 aa consisting of a canonical tripartite structure: short, positively charged n-region, a central hydrophobic h-region, and a more polar c-region with a cleavage site (26,27). Mouse Enam has an additional 5Ј-untranslated exon, which was detected within the human first intron but has not been identified in the transcripts (28).…”

“…The major secretory stage enamel proteins are amelogenin (21,22), ameloblastin (23)(24)(25), and enamelin (26,27). These proteins function specifically during enamel formation, and the disease phenotypes exhibited by mice lacking these genes are confined to the developing teeth and include enamel agenesis (28 -30).…”
