“…On the other hand, the LH/CG receptor truncated at residue 329 or further downstream is secreted to a limited extent (45,46). Alternately spliced truncated forms of TSHR mRNA have been detected in thyroid tissue (47,48); however, whether these transcripts are actually expressed and, if so, secreted by thyrocytes is unknown.…”

“…The presence of a thyroidal m-RNA encoding the ligand-binding region of the human TSHR, but not the transmembrane region, provides the potential for the production of soluble receptors, which could play an important role in autoimmune thyroid disease (24). It has also been reported that TPO can be detected in the blood of normal subjects and of patients with Graves' disease (25).…”

“…As discussed, the only study in which amplification of the full-length TSHR has been attempted is that of Feliciello et al (1993) in which the cell type containing the transcript has yet to be identified. We have demonstrated the presence of the 1.3-kb variant in Graves' thyroid, extra-ocular muscle and peripheral blood mononuclear cells and to a lesser extent in fat and fibroblasts using PCR primers specific for the exon 1-8 variant described by Graves et al (1992) in RTPCR experiments followed by Southern blotting and sequencing . Furthermore, we have measured TSH mediated protein synthesis, cAMP and glycosaminoglycan (GAG) production in cultured fibroblasts.…”

“…To add to the complexity, splice variants of variable lengths have been found for the TSH (Graves et al, 1992) (Koo et al, 1994). A summary of these variants is shown in Fig.…”
