Cloning and Expression of Two Carboxylesterases, and Their Activity Modulation in Chinese Mitten crab Eriocheir sinensis under Pesticide Exposer

Abstract: Carboxylesterases (CXEs) belong to a family of multifunctional enzymes. They metabolize drugs, environmental toxicants, and carcinogens, and inhibit bacterial pathogenesis. In this study, the full-length cDNAs of ES-CXE3 (2,444 bp) and ES-CXE4 (2,385 bp) were cloned from the Chinese mitten crab, Eriocheir sinensis. Sequence analysis showed that both ES-CXE sequences contained the catalytic triplet structure characteristic of the CXEs superfamily. Alignment and phylogenetic analyses revealed that the two ES-CXE… Show more

“…We assumed that M. rosenbergii may mitigate the toxicity of intracellular Cu accumulation by sequestering, utilizing, and excreting excess Cu. In addition, JHE-like carboxylesterase 2 (CXE2) and cytochrome P 450 (CYP 450) are involved in detoxification and protection against oxidative damage in crustaceans [63,64]. Cu exposure also induced the expression of CYP450 (CL1886.Contig2_All) and CXE2 (CL426.Contig5_All).…”

“…The activities of hepatopancreatic immune‐related enzymes are measured, and the immune‐related enzymes include catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA), acid phosphatase (ACP), alkaline phosphatase (AKP) and lysozyme (LZM) were measured using specific assay kits based on the manufacturer's protocol (Nanjing Jiancheng, China), two detoxification enzymes (CarEs and GSTs) in the hepatopancreas were measured as well. The method measured and activity calculation was performed according to the Zang, et al (Zang, Zhang, Huang, Ame, & Shen, 2020). Data were statistically analysed with SPSS v.18.0 using single‐factor ANOVA and Duncan's multiple range test, considering p < 0 .…”

“…For instance, some authors report evident seasonal fluctuation in acetylcholinesterase (AChE) (e.g., Menezes et al, 2006 ), a key enzyme in cholinergic signal transmission in the sensory and neuromuscular systems that therefore appears to be likely related to animals’ locomotor activity ( Anita et al, 2017 ). Moreover, carboxylesterases (CbE), a multifunctional group of enzymes relevant in lipid synthesis and decomposition and signal transmembrane transduction ( Zang et al, 2020 ), are involved in the metabolism of endogenous compounds ( Nos et al, 2021 ), and thermal stress can compromise their capability to face environmental chemical stressors ( Saiz et al, 2022 ).…”
