Cloning and expression of the gene for the Avi-3 antigen of Mycobacterium avium and mapping of its epitopes

Abstract: The Avi-3 antigen, which is found only in Mycobacterium avium culture sonic extracts, is species specific and results in strong skin test activity in guinea pigs sensitized with heat-killed M. avium. Its gene was cloned by using a previously developed single-probe method and was sequenced. The gene encoded a 194-amino-acid polypeptide with a molecular weight of 21,500. A recombinant Avi-3 antigen expressed in Escherichia coli reacted with monoclonal and polyclonal antibodies raised against the native Avi-3 ant… Show more

“…oxyR lesions and Mycobacterium tuberculosis susceptibility to isoniazid 891 organic peroxides to their corresponding alcohols (Christman et aL, 1985;Tartaglia et aL, 1990;Mathieu et aL, 1992;Chae et aL, 1994). Homologues of ahpC have been characterized in several bacteria, including Mycobacterium avium (Yamaguchi et aL, 1992) and M. leprae (MycDB cosmid B38) (Bergh and Cole, 1994). In M.…”

“…The gene was designated ahpC because of its similarity to ahpC genes in other bacterial genera. M. bovis ahpC had highest homology to an ahpC-like gene from Mycobacterium leprae (GenBank Accession Number L01095) and to a gene encoding the Avi-3 antigen from Mycobacterium avium (Yamaguchi et al, 1992). In E. coli and S. typhimurium, AhpC, together with the flavin adenine dinucleotide (FAD)-containing protein AhpF, have been proposed to constitute alkyl hydroperoxide reductase .…”

“…The 38-kDa antigen from M. tuberculosis was shown to have two possible epitopes (36); one of these has been found to be specific (35). In vitro studies of a 21.5-kDa Mycobacterium avium antigen identified two epitopes when recombinant fusion proteins and synthetic peptides were used (39).…”
