In order to investigate the effects of protein kinase C (PKC) expression on cellular growth and morphology, we established mouse fibrobiast cell pop~ations which expressed the rat pkc-y gene under the control of a retroviral promoter. NIH 3T3 stable ~nsf~tants displayed a three-fold increase in total PKC levels. These cells appeared mo~holo~~lly unaltered but exhibited a stronger mitogenic response to 12~0)-tetradecanoylphorbol-13-acetate (TPA) and cardiolipin (CL) as well as enhanced growth in semisolid medium in the presence of TPA. Thus, at these enzyme levels, PKC conferred growth advantages to NIH 3T3 cells only in response to specific activators.

It is known that PKC is differently expressed in brain and the peripheral nervous system and is involved in cellular differentiation. We have analyzed 9 human neuronal‐derived crest‐cell lines for PKC‐α mRNA. Seven out of nine expressed 9.0 kb and 4.0 kb PKC‐α mRNAs but three had high level of 9.0 kb transcription. The different expression of the two messenger RNAs may result from alternative splicing and a different degree of cell maturation. The same cell lines were studied for MYCN gene expression. A possible relation between the two genes is discussed. One cell line expressing high levels of both PKC‐α mRNA was treated with 10−1 M retinoic acid (RA). The expression of both messenger RNAs was suppressed when the cells achieved a morphological differentiation and showed neurite‐like processes. A decrease of PKC‐α gene expression was associated to down regulation of MYCN mRNA. These preliminary results suggest that PKC suppression of PKC‐α mRNA is associated with reversion of the malignant phenotype.

AbstractThe transcript levels of the protein kinase C (PKC) isoform genes during the development of a kindled epileptogenic focus, elicited by stimulation of Schaffer collateral/commissural fibres in the CA1 area of the rat hippocampus, were compared with the expression levels in control animals using a semi-quantitative in situ hybridization approach. In the hippocampus of control animals, the levels of PKC-o~-s r transcripts showed a gene-specific expression pattern and significant differences in expression level were observed between the neurons of CA1, CA3 and fascia dentata. In the early stages of kindling epileptogenesis, i.e. following 6 and 14 afterdischarges, specific changes in the expression levels of PKC-/3, -e, and -5 but not of PKC-a, -Y, and -6 were found. PKC-/3 expression was decreased in CA1, while the PKC-e and _r specific hybridization signals were increased in CAI, CA3 and fascia dentata. In fully kindled animals, that had experienced 10 generalized seizures, most expression levels tended to return to control values. One month after the last seizure no significant alterations were encountered. These results indicate an involvement of specific PKC-isoform gene expression in the induction of an epileptogenic focus, but not in the maintenance of the long-lasting kindled state.
