Cloning and expression of chitinase A, B, and C (chiA, chiB, chiC) genes from Serratia marcescens originating from Helicoverpa armigera and determining their activities

Abstract: IntroductionChitin, a linear polymer of β-1,4-linked N-acetyl-Dglucosamine (GlcNAc), is one of the most abundant biopolymers in nature. It is the main component of the exoskeleton of many invertebrates such as crustaceans, insects, and spiders and a structural component of the cell walls of most fungi and algae. In insects it functions as scaffold material, supporting the cuticles of the epidermis and trachea as well as the peritrophic membrane (PM) lining the gut epithelium (Merzendorfer and Zimoch, 2003;Ruiz… Show more

“…Also, domain positions were opposite in S. marcescens-ChiC and viral chitinases, while the glyco_18 and GH_18 domains located at the C terminal of viral chitinases showed similar domains located at the N terminal of S. marcescens-ChiC. S.marcescens-ChiA, structurally analyzed previously by Danişmazoğlu et al (2015), had a similar domain organization to those of AcNPV-Chi and CpGV-Chi.…”

“…Bacterial chitinase protein (chitinase C) belongs to Serratia marcescens that was previously isolated from dead Helicoverpa armigera (Hubner, 1808) larvae. The S. marcescens chitinase gene (accession number: KF823632) was provided cloned into the pET-28a (+) expression vector (Danişmazoğlu et al 2015). Labreared culture of Galleria mellonella (Linnaeus, 1758).…”

“…Among hydrolytic enzymes, chitinolytic enzymes are the most important for the biocontrol of fungal phytopathogens. Chitinases are encoded by a number of genes that include, e.g., chiA , chiB , chiC and chiD , chiF , chiG , chiR , chiW , and chiX ( Alam et al, 1996 ; Hamilton et al, 2014 ; Danişmazoğlu et al, 2015 ; Martínez-Zavala et al, 2020 ; Azizoglu et al, 2021 ; Kumar et al, 2022 ). The efficacy of chitinases produced by B. pumilus against fungi has been documented in a few studies.…”
