Cloning and endonuclease restriction analysis of uidA and uidR genes in Escherichia coli K-12: determination of transcription direction for the uidA gene.

Abstract: The two genes of the Escherichia coli K-12 uid region (the structural gene uidA and the regulatory gene uidR) were isolated on a ColE1-uid hybrid plasmid from the bank of Clarke and Carbon. We made a restriction map of this region and correlated it with the genetic map by subcloning the uid restriction fragments into plasmids pBR322, pBR325, and pACYC177. In these plasmids, amplification of the products of the uidA and uidR genes occurred. The enzyme coded for by uidA was identified by polyacrylamide gel elect… Show more

“…Restriction targets for HindlIl (H), EcoRI (R), and PstI (P) and the fragment sizes (in kilobases) are indicated. Genetic symbols: terC, terminus of chromosome replication; g48, unidentified gene encoding 48-kilodalton product; fumA fumarase; manA, mannosephosphate isomerase; uidAoR, ,B-glucuronidase operon (4). The relative orientation of the physical and genetic maps is derived from Bouche (6).…”

“…The UidR regulon ( Table 2) allows for the growth of E. coli on the D-glucuronate and D-galacturonate moieties of the ␤-glucuronides and ␤-galacturonides present in mucus (10). After entering the cell, glucuronate and galacturonate are degraded via parallel pathways (Fig.…”

“…When plasmids pREG8 and pREG9 were introduced into strain 2510 (lacZY deletion) and into strain HB101 (lacY mutation), a Lac' phenotype on lactose-containing MacConkey agar plates was restored only in the transformants of strain HB101 (6). This indicates that the BamHI fragment cloned into pMC874 possessed a promoter activity in the left and right directions When plasmid pRE2 carrying the exuR regulatory gene was added in trans in these transformants, 3-galactosidase synthesis was strongly reduced, suggesting that transcription of the two fused genes was regulated like transcription of the original exuT and uxaA genes, by the ExuR repressor ( Table 2).…”
