Cloning and characterization of theHpallmethylase gene

Abstract: The HpaII restriction-modification system from Haemophilus parainfluenzae recognizes the DNA sequence CCGG. The gene for the HpaII methylase has been cloned into E. coli and its nucleotide sequence has been determined. The DNA of the clones is fully protected against cleavage by the HpaII restriction enzyme in vitro, indicating that the methylase gene is active in E. coli. The clones were isolated in an McrA-strain of E. coli; attempts to isolate them in an McrA+ strain were unsuccessful. The clones do not exp… Show more

“…We first turned to the HpaII C5-MTase (M.HpaII), which recognizes C C GG targets in DNA (27) and which thus can potentially be useful to study mammalian DNA modification at CpG sites. As expected, the WT enzyme showed low or no activity with AdoMet analogs 2 and 4 , respectively.…”

“…SmaI endonuclease protection assay. Expression and activity of the recombinant M.SpyI was tested using a combination of protocols described previously (11,29) following the instructions for the pBADTOPO TA Expression Kit. Briefly, recombinant M.SpyI expression was induced by the addition of 0.2% or 2% L-arabinose to cultures at an optical density at 600 nm of 0.5, and growth then continued for 2 or 4 h at 37°C, respectively.…”
