Cloning and characterization of the human glutathione synthetase 5′-flanking region

Abstract: GSH synthesis occurs through a two-step enzymatic reaction driven by GCL (glutamate-cysteine ligase; made up of catalytic and modifying subunits) and GSS (glutathione synthetase). In humans, oxidative stress regulates GCL expression in an antioxidant response element-dependent manner via Nrf2 [NFE (nuclear factor erythroid)-related factor 2]. In the rat, GSS and GCL are regulated co-ordinately by oxidative stress, and induction of GSS further increases GSH synthetic capacity. Transcriptional regulation of the … Show more

“…The promoter of the GSH synthetase gene contains multiple binding sites for STAT family members including STAT5, suggesting that STAT5 might regulate the transcriptional activation of this gene. 42 Moreover, the synthesis of GSH or the activity of antioxidant enzymes such as GSH reductase and GSH peroxidase are modulated by the Hsp27 and Hsp70. Thus, it is likely that STAT5 modulates expression or activity of these enzymes via an indirect effect on Hsp27 and Hsp70 expression.…”

“…Accumulating evidence shows that Nrf2 activation can consequently elevate the expression and enzymatic activities of ␥-glutamyl-cysteine synthetase/GST as well as the GSH level in cells (56,57). Considering the reversible nature of sulfhydryl modification by sulforaphane, the elevated GSH levels may favor the restoration of NES TA activity and trigger Nrf2 nuclear export.…”

“…The rat GCLM promoter also has a functional ARE element (−295 to −285) (Yang et al, 2005b). This ARE element is important for basal promoter activity as well as TNFα-mediated induction of rat GCLM (Yang et al, 2005). AP-1, NFκB and Nrf2 are all positive regulators of the rat GCLM gene and are induced by TNFα treatment (Yang et al, 2005b).…”

“…The gene encoding GS was cloned in 1995 (Huang et al, 1995) but its transcriptional regulation has only been characterized recently after cloning of the rat and human GS promoters Lee et al, 2005). 1,10-phenanthroline, a metal chelating agent, was reported to induce GS expression but the mechanism was not examined (Sun, 1997).…”
