Cloning and Characterization of Rat Density-Enhanced Phosphatase-1, a Protein Tyrosine Phosphatase Expressed by Vascular Cells

Abstract: We have cloned from cultured vascular smooth muscle cells a protein tyrosine phosphatase, rat density-enhanced phosphatase-1 (rDEP-1), which is a probable rat homologue of DEP-1/HPTPη. rDEP-1 is encoded by an 8.7-kb transcript and is expressed as a 180- to 220-kD protein. The rDEP-1 gene is located on human chromosome 11 (region p11.2) and on mouse chromosome 2 (region 2E). “…hDEP-1 was isolated separately from a human myeloid cell line and referred to as HPTPh [13]. A rat Dep-1 homolog (rDEP-1) was cloned recently and was also found to be regulated in response to density-dependent inhibition of growth in vascular cells, although curiously not in fibroblasts [14]. In this study we show that mDEP-1 is indeed a marker for macrophage differentiation and that the expression is highly regulated in mature macrophages.…”
