Cloning and characterization of mouse Klk27, a novel tissue kallikrein expressed in testicular Leydig cells and exhibiting chymotrypsin‐like specificity

Abstract: A cDNA clone of a new mouse tissue kallikrein, designated mKlk27, was isolated from an adult mouse testis cDNA library. mKlk27 was expressed in the submaxillary glands and testis of the mouse. In testis, mKlk27 gene was expressed exclusively in the Leydig cells of the adult mouse. Active recombinant mKlk27 exhibited chymotrypsin-like cleavage specificity. A single amino-acid substitution of Gly for Asp at position 209 in mKlk27 resulted in complete loss of its chymotryptic activity but acquisition of tryptic a… Show more

“…These values are larger than those calculated from the amino acid sequences of these proteins. Such differences in molecular size between the theoretical and experimental values have previously been reported for other proteins, including serine proteinase family members [11,12,18,36]. Interestingly, the testicular tissues in the present study contained Gzmn in two forms (M r ϭ 31 kDa and 30 kDa).…”

“…At present, 11 Gzm genes have been identified for the mouse, including granzyme L (Gzml), which is presumed to be a pseudogene with early termination sites [3]. Our current data regarding RT-PCR analysis for the 10 functional Gzm species revealed that in addition to Gzmn and Gzmb, four other genes (Gzma, Gzmc, Gzmk, and Gzmm) are expressed in the mouse testes (Figs.…”

“…Kallikrein 24 has been shown to be androgen but not estrogen regulated in vitro (45), and the expression of these three kallikreins has also been found to be reduced in the testis of AR mutant mice (42). In the testis, kallikreins 21, 24, and 27 are exclusively expressed in the Leydig cells, and their expressions are shown to increase during development, being highest at the age of 8 wk (45)(46)(47). Thus, they can be considered as adult-type Leydig cell markers.…”
