Cloning and characterization of different human sequences related to the onc gene (v-myc) of avian myelocytomatosis virus (MC29).

Abstract: We have studied the genomic organization of human cellular sequences (c-myc) homologous to the transforming gene (v-myc) of avian myelocytomatosis virus (MC29). Southern blotting experiments using v-myc probes showed that several fragments of the human genome contain sequences related to the central part of v-myc but only few of them are homologous to the 3' portion of the viral gene. Several recombinant phages which represent different regions of the genome containing c-myc-related sequences were isolated fro… Show more

“…The restriction map of XhR1-15 shown in Fig. 1 appears nearly identical to that described by Dalla-Favera et al (12) and is consistent with the human c-myc nucleotide sequence (2,9). Additional restriction sites in the human genome surrounding the cloned fragment were determined and are also shown in Fig.…”

“…1). These other sequences, which have homology to the 5' half of v-myc, have been suggested to be pseudogenes (12). Only RNA transcribed from the full-length locus will be detected with the ClaI-EcoRI probe.…”

“…To construct the MBP/c-myc transgene, a KpnI ± XbaI fragment containing the 3 kb MBP promoter was inserted in the 5' orientation at the KpnI ± XbaI site of a genomic clone of the human c-myc gene (Dalla-Favera et al, 1982). In order to insure high levels of expression of the c-myc transgene, exon 1, which contains negative regulatory elements, was removed (Xu et al, 1991).…”

“…Vimentin probe was kindly provided by R Liem. c-MYC exon 3 and exon 1 probes were obtained from the pMC41RC and pMC41ER plasmids, respectively (Dalla-Favera et al, 1982). The SV40 poly(A) tail probe and human b-actin probe were previously described (Wu et al, 1999b).…”
