Cloning and characterization of a novel endothelin receptor subtype in the avian class

Abstract: Endothelin 3 (EDN 3) and the endothelin receptor B (EDNRB) are involved in the development of neural crest and particularly of the melanocytes and the enteric nervous system. We reported previously that the avian EDNRB gene is expressed in the neural fold before crest cell migration and later on in all the neural crest derivatives except, at any developmental stage, in the melanocytic lineage. However, quail melanoblasts proliferate in response to EDN 3 stimulation in vitro. These observations prompted us to s… Show more

“…A paralogue of EDNRB (designated EDNRB2), found to be specific to the melanocytic lineage, was cloned in quail by Lecoin et al (1998). A mutant in quail, which had an amino acid substitution and reduced gene expression in the ENDRB2 gene, displayed white plumage with wild-type colored spots (Miwa et al 2007).…”

“…Figure 4a illustrates the RT ± PCR analysis that we performed on RNA from tsMyb/EGFR cells grown at both 378C and 428C. This analysis detected expression of tyrosinase (1006) which is required for the synthesis of melanin, the slug zinc ®nger protein which is expressed on migrating neural crest cells (Nieto et al, 1994), the bHLH protein Mitf which is essential for melanocyte di erentiation (Opdecamp et al, 1997), the endothelin receptor (EDNRB2, Lecoin et al, 1998), melanotransferrin (EOS47, McNagny et al, 1996, the chicken homologue of the melanoma associated antigen MUC18 (HEMCAM, Vainio et al, 1996) and c-Myb. We also consistently detected the expression of HEMCAM using the antibody JS3 (not shown).…”
