Cloning and Characterization of a Probenazole-Inducible Gene for an Intracellular Pathogenesis-Related Protein in Rice

Abstract: Probenazole (3-allyloxy-1,2-benzisothiazole-1,1-dioxide) induces disease resistance in rice against rice blast fungus. To investigate the molecular mechanism of probenazole-induced resistance, we isolated and characterized a cDNA clone of a probenazole-inducible gene in rice, which encoded a protein designated PBZ1. Sequence analysis revealed that significant homology at the amino acid level exists between the predicted PBZ1 protein and intracellular pathogenesis-related (IPR) proteins. Accumulation of PBZ1 mR… Show more

“…C, HPLC analysis of reaction products after the microsomal SL proteins were assayed for enzyme activity using tryptamine as a substrate. from fungal pathogens (25). 6 Expression of these defense genes was clearly induced at 12 h after serotonin treatment, but no induction was found with tryptamine ( Fig.…”

“…These genes include those for 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) (Nelson et al 1994), lipoxygenase (Peng et al 1994), phenylalanine ammonia-lyase (PAL) and chitinase (Xu et al 1996). Transcripts of some other rice genes, such as PBZ1 (Midoh and Iwata 1996), RMa1 and RMa3 , accumulate in response to infection by M. grisea. Likewise, transcription of rice genes for HMGR, chitinase and GDP-dissociation inhibitors (GDIs) is induced by elicitors derived from M. grisea cell wall (Nelson et al 1994;Xu et al 1996;Kim et al 1999).…”

“…They were first identified in parsley (Petroselinum crispum; Somssich et al, 1988) and later isolated from several plant species and tissues (Matton and Brisson, 1989;Crowell et al, 1992;Warner et al, 1994;Breda et al, 1996;Midoh and Iwata, 1996;Huang et al, 1997;Lo et al, 1999;Yu et al, 2000;Wu et al, 2003). PR-10 proteins are typically small (from 155-163 residues) cytoplasmic proteins with acidic pI (Liu and Ekramoddoullah, 2006).…”
