Cloning and characterisation of a zona pellucida 3 cDNA from a marsupial, the brushtail possum Trichosurus vulpecula

Abstract: The mammalian zona pellucida (ZP) is an extracellular glycoprotein coat that plays vital roles throughout fertilisation and preimplantation development. Like that of eutherian mammals the brushtail possum ZP is composed of three glycosylated proteins of 137 kDa, 92 kDa and 62 kDa. The 62 kDa protein is a ZP3 orthologue based on its nucleotide and deduced amino acid sequence. The brushtail possum ZP3 cDNA isolated in this study is 1305 nucleotides with an open reading frame encoding a 422 amino acid peptide of … Show more

“…The conservation of the ZP domain in such evolutionary divergent species as teleost fish, amphibians, birds, marsupials (Mate and McCartney, 1998;McCartney and Mate, 1999), and mammals suggests that not only has this protein domain been duplicated in vertebrates but that it has been conserved and used as an egg envelope protein in species that diverged several million years ago.…”

“…The zona pellucida glycoconjugate content of several marsupial species was investigated using differential lectin histochemistry [Chapman et al, 2000;Breed at al., 2002] to show interspecific variation in glycoconjugate content, with mannose-containing glycoconjugates exhibiting the greatest variation. Considerable homology exists between the three marsupial zona proteins, ZP1, 2 and 3, and those in eutherians [Mate and McCartney, 1998;McCartney and Mate, 1999;Haines et al, 1999] suggesting that the carbohydrate component might be significant for cell-zona attachment.…”

“…A number of recent studies have established that ZPA (= mouse ZP2) [Mate and McCartney, 1998], ZPB (= mouse ZP1) [Haines et al, 1999] and ZPC (= mouse ZP3) [McCartney and Mate, 1999] are present in marsupials. A summary of the sequence identity of the nucleic acids and amino acids of the three zona proteins is shown in table 2.…”
